Are you using FBA? If so, Prime eligibility is automatic unless the item itself is not eligible.
If offering FBM then you have to be enrolled in Seller Fulfilled Prime -- see this page:

I want to confirm what you learned from bookwormapril regarding Prime.
- If you are using Fulfilled by Amazon (FBA) then your products will be available to Amazon Prime Customers.
- If you are fulfilling your own orders, known as Merchant Fulfilled Network (MFN) then you would need to apply for and be approved for Seller Fulfilled Prime.
